R21: Catalytic Tool & Technology Development in Kidney, Urologic, & Hematologic Diseases (Clinical Trial Not Allowed)

The catalytic tool and technology development in kidney, urologic, and hematologic diseases r21 at niddk.

NIDDK funds catalytic tool and technology development in kidney, urologic, and hematologic diseases through the R21 grant mechanism. The R21 provides funding of approximately $125,000 per year for 2 years. Strong applications will develop new tools or technologies in kidney, urology or hematology with broad utility beyond the applicant's own laboratory and ideally push the entire field forward. As such, applications are required to provide plans for making the proposed tool or technology available to the national research community.

  • The R21 is not intended to support the improvement of tools or technologies that have already been deployed in kidney, urologic, or hematologic science. Extensive preliminary data demonstrating feasibility is an indication that the proposal is beyond the scope of this program.
  • The R21 is not intended to support “mini-R01s” with a traditional hypothesis-driven research plan.

Prospective applicants are strongly encouraged to contact:

Find a Funding Opportunity

stage indicator - find a funding opportunity

Comparing Popular Research Project Grants—R01, R03, and R21

This page discusses the three most common types of Research Projects (R) : the NIH standard independent R01, the smaller R03, and the exploratory/developmental research R21 activity codes.

For other types of grants, check Types of Funding Opportunities .

We strongly advise you to contact an NIAID program officer before you start to develop your application. He or she can advise you on whether your proposed project is appropriate for a particular grant type.

The main driver of your decision for one activity code over another should be the scientific scope of the work being proposed. Normally, R01-type projects are mature ideas with preliminary data supporting a hypothesis. R21 projects are usually high-risk/high-reward and require a high degree of innovation. R03 projects are usually descriptive and are used to generate hypotheses and data to support them.

Is an R01 Right for You?

An R01 is for mature research projects that are hypothesis-driven with strong preliminary data. R01s provide up to five years of support, with a budget that reflects the costs required to complete the project.

If you have strong preliminary data supporting your hypothesis, we recommend applying for an R01. For more on qualifying for an R01, refer to Determine Eligibility for NIAID Grants .

New investigators, when trying to determine whether submitting an R01 application is a good idea, consider the following:

  • Do you have strong preliminary data that support your hypothesis/hypotheses?
  • Is there a strong rationale for the proposed study?
  • Do you have a proven track record of experience in the field, including publications in scientific journals?
  • Do you or your collaborators have sufficient expertise to accomplish the goals of the proposed work?

Read Know Your Audience and Write Your Research Plan for more information about writing your application.

Reviewers are asked to consider that new investigators may have a more modest track record and publication record than established investigators. Peer reviewers are advised to look more at your potential than your past achievements. Also note that NIAID new investigators benefit from a more favorable payline for R01 applications.

For additional details, check Information for New Investigators .

Smaller Awards (R03, R21)

If your project is not ready for an R01, you can consider a two-year small grant (R03) or exploratory/developmental research grant (R21) to generate preliminary data or develop approaches that could support a hypothesis that can be tested under the R01 activity code. If you are considering either of these activity codes, get more advice from a program officer before applying.

R03 Is Small

Don't let the word "small" fool you into thinking that an R03 is especially easy to get; it's not. Like most other grant activity codes, your application will have stiff competition, will undergo peer review, and must be suitable for the activity code.

Your project must be tightly focused, able to be completed in two years, and manageable with a maximum of $100,000 in direct costs over the two-year period. If your project doesn't meet these criteria, it's not likely to be well received by reviewers.

Examples of projects that fit the R03 include:

  • Pilot or feasibility studies
  • Descriptive studies
  • Secondary analysis of existing data
  • Small, self-contained research projects
  • Adaption or further development of research methodology or new research technology

To see the type of projects that have been funded, go to NIH RePORTER and select R03 in the Activity Code field. Also refer to R03 Sample Applications and Summary Statements .

New Investigators: R03 Might Not Be Your Best Option

At first glance, the R03 seems well suited to a new investigator. You can request up to $50,000 a year in direct costs and do not need preliminary data. And for someone who's never written a grant application, the shorter six-page Research Strategy (versus 12 pages for an R01) might be appealing.

That said, however, the smaller page limit means you need a concise, tightly focused research plan. The short time provided to complete the project and the limited budget mean you have little room for course correction.

Thus, when considering the R03 activity code, be sure to think about the scope of the research you are proposing, paying special attention to the timeline of the project and the funds that will be needed to complete the project.

Should You Apply for an R21?

The R21 activity code is intended to encourage exploratory/developmental research by providing support for the early and conceptual stages of project development. NIH has standardized the exploratory/developmental grant (R21) application characteristics, requirements, preparation, and review procedures in order to accommodate investigator-initiated (unsolicited) grant applications.

R21 projects are usually highly innovative driving the high-risk/high-reward principle that is characteristic of this activity code. R21 awards are intended to allow short duration funding (two years) for innovative ideas. R21 projects should be distinct from those supported through the traditional R01 and are not intended as a bridge between training and obtaining an R01 grant.

Here we lay out some considerations you'll want to pay attention to if you're thinking about submitting an R21 application.

R21 Pros and Cons

  • Allows you to introduce novel scientific ideas, model systems, tools, agents, targets, and technologies that have the potential to substantially advance biomedical research.
  • Preliminary data are not required for an R21 application.
  • If you're applying for an R21 to generate preliminary data, you could end up coming up short for time with a two-year grant.
  • In addition, the purpose of the R21 activity code is not necessarily to support hypothesis-based research.
  • Further, your R21 won't benefit from our higher R01 payline for new and early-stage investigators.

For a comparison of several key characteristics of R21s and R01s, see the next section.

R21 or R01?

This table compares some features of the parent R21 and R01 fund opportunity announcements (NOFOs). Note that institutes may publish their own funding opportunities (initiatives) using the R21 or R01 activity codes with different characteristics.

* Data come from RePORT's Research Project Success Rates for NIAID .

No matter which funding opportunity you choose, you are advised to discuss with your program officer to make sure you are applying to the appropriate NOFO. Also, you need to consider where your financial support will come from should you have a funding gap while awaiting receipt of an NIH award. Read more in Approaches for Staying Funded .

To find information about different activity codes, go to NIH's Types of Grant Programs page.

Part 2. Full Text of Announcement

Section i. funding opportunity description.

The BRAIN Initiative

The B rain R esearch through A dvancing I nnovative N eurotechnologies (BRAIN) Initiative is aimed at revolutionizing our understanding of the human brain. By accelerating the development and application of innovative technologies, researchers will be able to produce a new dynamic picture of the brain that, for the first time, will show how individual cells and complex neural circuits interact in both time and space. It is expected that the application of these new tools and technologies will ultimately lead to new ways to treat and prevent brain disorders.

NIH is one of several federal agencies involved in the BRAIN Initiative. Planning for the NIH component of the BRAIN initiative is guided by the long-term scientific plan, " BRAIN 2025: A Scientific Vision ," which details seven high-priority research areas and calls for a sustained federal commitment of $4.5 billion over 12 years. This FOA and other recently issued FOAs are based on careful consideration by the NIH of the recommendations of the BRAIN 2025 Report, and input from the NIH BRAIN Multi-Council Working Group. Videocasts of the NIH BRAIN Multi-council Working Group are available at http://www.braininitiative.nih.gov/about/mcwg.htm .

In addition to the National BRAIN initiative, the NIH continues to have a substantial annual investment in neuroscience research. The Institutes and Centers contributing to the NIH BRAIN Initiative ( http://braininitiative.nih.gov/ ) support those research efforts through investigator-initiated applications as well as through specific FOAs. Potential applicants to this FOA are strongly encouraged to contact Scientific/Program staff if they have any questions about the best FOA for their research.

To enable rapid progress in development of new technologies as well as in theory and data analysis, the BRAIN Initiative encourages collaborations between neurobiologists and scientists from statistics, physics, mathematics, engineering, and computer and information sciences; and NIH welcomes applications from investigators in these disciplines.

NIH encourages BRAIN Initiative applications from investigators that are underrepresented in the biomedical, behavioral, or clinical research workforce (see data at http://www.nsf.gov/statistics/showpub.cfm?TopID=2&SubID=27 and the most recent report on Women, Minorities, and Persons with Disabilities in Science and Engineering ). Such individuals include those from underrepresented racial and ethnic groups, those with disabilities, and those from disadvantaged backgrounds.

The BRAIN Initiative will require a high level of coordination and sharing between investigators. It is expected that BRAIN Initiative awardees will cooperate and coordinate their activities after awards are made by participating in Program Director/Principal Investigator (PD/PI) meetings and in other activities.

This FOA is related to the recommendations in sections II.2, II.3, and II.4 from the BRAIN 2025 Report. These three recommendations call for accelerated development of new large-scale recording technologies and tools for neural circuit manipulation. These new technologies and approaches will provide unprecedented opportunities for exploring how the nervous system encodes, processes, utilizes, stores, and retrieves vast quantities of information. A better understanding of this dynamic neural activity will enable researchers to seek new ways to diagnose, treat, and prevent brain disorders.

Achieving these goals requires the ability to record simultaneously from thousands or tens-of- thousands of neurons contributing to the dynamic activity in a neural circuit. The relevant activity may be in clusters of cells packed closely together or may be in widely distributed circuits. Current microelectrode and imaging technologies are limited in the number of cells from which activity can be isolated and sampled simultaneously, by the size or location of the area to be sampled, by the depth of penetration, and by the invasiveness of the technique that might prohibit their use in human experimentation. Non-invasive technologies suitable for use in humans are currently limited in spatial resolution and temporal dynamics, as well as in their reflection of on-going electrical activity in circuit elements. This FOA seeks entirely new ideas, concepts and/or approaches from physics and engineering, and biology, for how these limitations might be overcome to enable increased recording capabilities on the scale of one or more orders of magnitude beyond that of current technology.

This FOA is also related to the goals of the updated BRAIN 2.0 Report, which recommends expanding the functionality and integration of electrophysiological and neurochemical methods. The next generation of recording/modulation technology development should include efforts to improve cell-type and neurochemical specificity. Thus, an equally important goal of this FOA is to discover novel ideas for technology capable of precisely manipulating activity in circuits. Dissecting the function of neural circuits requires the ability to precisely activate or inactivate brain cells (both neuronal and non-neuronal) in order to investigate underlying mechanisms and demonstrate causality. Current technologies such as microstimulation and optogenetic approaches are limited in specificity, temporal dynamics, and by the invasiveness of the technique.

Applications are expected to propose the development of ideas in the earliest stages for entirely new approaches for neural recording and/or precise manipulation of neural activity. Such ideas could encompass unique and innovative combinations of existing technology that create a synergistic result. An important goal is to stimulate new thinking and concepts for accelerating development of novel technologies that break current barriers to neural recording and/or manipulation. In addition to experimental approaches, this FOA may support early-stage testing using calculations, simulations, computational models, or other mathematical techniques for demonstrating that the signal sources and/or measurement technologies are theoretically capable of meeting future demands. The support might also be used for building and testing phantoms, prototypes, in-vitro or other bench-top models in order to validate underlying theoretical assumptions in preparation for companion FOAs aimed at proof-of concept testing in animal models.

Applications are expected to propose research that will explore ideas in their earliest stages of development in order to be responsive to goals and objectives of this FOA. The following topics are non-responsive: i) further development of existing technology; ii) neurobiological hypothesis-testing; iii) validation and/or refinement of current technology; or iv) development of analytical methods to be applied to existing technology and/or data.

The technologies that would ultimately evolve from these new approaches should be compatible with experiments in humans and/or behaving animals and should dramatically improve capabilities in order to enable experiments that are currently not possible. Preliminary data are not required; however, they may be included if available.

Applications from individuals not usually associated with neuroscience research or teams that cross boundaries forming interdisciplinary collaborations capable of bringing new and untested ideas are particularly encouraged. Accordingly, applicants might consider, where appropriate, multi-PD/PI applications that integrate appropriate expertise, including but not limited to biological, chemical and physical sciences, engineering, computational modeling and statistics.

Section V. Application Review Information

1. criteria.

Note: Effective for due dates on or after January 25, 2023, the Data Sharing Plan and Genomic Data Sharing Plan (GDS) as part of the Resource Sharing Plan will not be evaluated at time of review.

Only the review criteria described below will be considered in the review process. Applications submitted to the NIH in support of the NIH mission are evaluated for scientific and technical merit through the NIH peer review system.

For this particular announcement, note the following:

The R21 exploratory/developmental grant supports investigation of novel scientific ideas or new model systems, tools, or technologies that have the potential for significant impact on biomedical or biobehavioral research. An R21 grant application need not have extensive background material or preliminary information. Accordingly, reviewers will emphasize the conceptual framework, the level of innovation, and the potential to significantly advance our knowledge or understanding. Appropriate justification for the proposed work can be provided through literature citations, data from other sources, or, when available, from investigator-generated data. Preliminary data are not required for R21 applications; however, they may be included if available.

The novel technology sought in this FOA is expected to be at the earliest stage of development, prior to empirical studies of proof-of-concept or preliminary data.

Reviewers will provide an overall impact score to reflect their assessment of the likelihood for the project to exert a sustained, powerful influence on the research field(s) involved, in consideration of the following review criteria and additional review criteria (as applicable for the project proposed).

Reviewers will consider each of the review criteria below in the determination of scientific merit, and give a separate score for each. An application does not need to be strong in all categories to be judged likely to have major scientific impact. For example, a project that by its nature is not innovative may be essential to advance a field.

Significance

Does the project address an important problem or a critical barrier to progress in the field? Is the prior research that serves as the key support for the proposed project rigorous? If the aims of the project are achieved, how will scientific knowledge, technical capability, and/or clinical practice be improved? How will successful completion of the aims change the concepts, methods, technologies, treatments, services, or preventative interventions that drive this field ?

Investigator(s)

Are the PD(s)/PI(s), collaborators, and other researchers well suited to the project? If Early Stage Investigators or those in the early stages of independent careers, do they have appropriate experience and training? If established, have they demonstrated an ongoing record of accomplishments that have advanced their field(s)? If the project is collaborative or multi-PD/PI, do the investigators have complementary and integrated expertise; are their leadership approach, governance and organizational structure appropriate for the project?

Does the application challenge and seek to shift current research or clinical practice paradigms by utilizing novel theoretical concepts, approaches or methodologies, instrumentation, or interventions? Are the concepts, approaches or methodologies, instrumentation, or interventions novel to one field of research or novel in a broad sense? Is a refinement, improvement, or new application of theoretical concepts, approaches or methodologies, instrumentation, or interventions proposed?

Are the overall strategy, methodology, and analyses well-reasoned and appropriate to accomplish the specific aims of the project? Have the investigators included plans to address weaknesses in the rigor of prior research that serves as the key support for the proposed project? Have the investigators presented strategies to ensure a robust and unbiased approach, as appropriate for the work proposed? Are potential problems, alternative strategies, and benchmarks for success presented? If the project is in the early stages of development, will the strategy establish feasibility and will particularly risky aspects be managed? Have the investigators presented adequate plans to address relevant biological variables, such as sex, for studies in vertebrate animals or human subjects?

If the project involves human subjects and/or NIH-defined clinical research, are the plans to address 1) the protection of human subjects from research risks, and 2) inclusion (or exclusion) of individuals on the basis of sex/gender, race, and ethnicity, as well as the inclusion or exclusion of individuals of all ages (including children and older adults) , justified in terms of the scientific goals and research strategy proposed?

Environment

Will the scientific environment in which the work will be done contribute to the probability of success? Are the institutional support, equipment and other physical resources available to the investigators adequate for the project proposed? Will the project benefit from unique features of the scientific environment, subject populations, or collaborative arrangements?

As applicable for the project proposed, reviewers will evaluate the following additional items while determining scientific and technical merit, and in providing an overall impact score, but will not give separate scores for these items.

Protections for Human Subjects

For research that involves human subjects but does not involve one of the categories of research that are exempt under 45 CFR Part 46, the committee will evaluate the justification for involvement of human subjects and the proposed protections from research risk relating to their participation according to the following five review criteria: 1) risk to subjects, 2) adequacy of protection against risks, 3) potential benefits to the subjects and others, 4) importance of the knowledge to be gained, and 5) data and safety monitoring for clinical trials.

For research that involves human subjects and meets the criteria for one or more of the categories of research that are exempt under 45 CFR Part 46, the committee will evaluate: 1) the justification for the exemption, 2) human subjects involvement and characteristics, and 3) sources of materials. For additional information on review of the Human Subjects section, please refer to the Guidelines for the Review of Human Subjects .

Inclusion of Women, Minorities, and Individuals Across the Lifespan

When the proposed project involves human subjects and/or NIH-defined clinical research, the committee will evaluate the proposed plans for the inclusion (or exclusion) of individuals on the basis of sex/gender, race, and ethnicity, as well as the inclusion (or exclusion) of individuals of all ages (including children and older adults) to determine if it is justified in terms of the scientific goals and research strategy proposed. For additional information on review of the Inclusion section, please refer to the Guidelines for the Review of Inclusion in Clinical Research .

Vertebrate Animals

The committee will evaluate the involvement of live vertebrate animals as part of the scientific assessment according to the following criteria: (1) description of proposed procedures involving animals, including species, strains, ages, sex, and total number to be used; (2) justifications for the use of animals versus alternative models and for the appropriateness of the species proposed; (3) interventions to minimize discomfort, distress, pain and injury; and (4) justification for euthanasia method if NOT consistent with the AVMA Guidelines for the Euthanasia of Animals. Reviewers will assess the use of chimpanzees as they would any other application proposing the use of vertebrate animals. For additional information on review of the Vertebrate Animals section, please refer to the Worksheet for Review of the Vertebrate Animal Section .

Reviewers will assess whether materials or procedures proposed are potentially hazardous to research personnel and/or the environment, and if needed, determine whether adequate protection is proposed.

Resubmissions

As applicable for the project proposed, reviewers will consider each of the following items, but will not give scores for these items, and should not consider them in providing an overall impact score.

Applications from Foreign Organizations

Reviewers will assess whether the project presents special opportunities for furthering research programs through the use of unusual talent, resources, populations, or environmental conditions that exist in other countries and either are not readily available in the United States or augment existing U.S. resources.

Select Agent Research

Reviewers will assess the information provided in this section of the application, including 1) the Select Agent(s) to be used in the proposed research, 2) the registration status of all entities where Select Agent(s) will be used, 3) the procedures that will be used to monitor possession use and transfer of Select Agent(s), and 4) plans for appropriate biosafety, biocontainment, and security of the Select Agent(s).

Resource Sharing Plans

Reviewers will comment on whether the following Resource Sharing Plans, or the rationale for not sharing the following types of resources, are reasonable: (1) Data Sharing Plan ; (2) Sharing Model Organisms ; and (3) Genomic Data Sharing Plan (GDS) .

Authentication of Key Biological and/or Chemical Resources:

For projects involving key biological and/or chemical resources, reviewers will comment on the brief plans proposed for identifying and ensuring the validity of those resources.

Budget and Period of Support

Reviewers will consider whether the budget and the requested period of support are fully justified and reasonable in relation to the proposed research.

2. Review and Selection Process

Applications will be evaluated for scientific and technical merit by (an) appropriate Scientific Review Group(s) convened by the National Eye Institute, in accordance with NIH peer review policy and procedures , using the stated review criteria . Assignment to a Scientific Review Group will be shown in the eRA Commons.

As part of the scientific peer review, all applications will receive a written critique.

Applications may undergo a selection process in which only those applications deemed to have the highest scientific and technical merit (generally the top half of applications under review) will be discussed and assigned an overall impact score.

Applications will be assigned on the basis of established PHS referral guidelines to the appropriate NIH Institute or Center. Applications will compete for available funds with all other recommended applications submitted in response to this FOA. Following initial peer review, recommended applications will receive a second level of review by the National Advisory Eye Council. The following will be considered in making funding decisions:

  • Scientific and technical merit of the proposed project as determined by scientific peer review.
  • Availability of funds.
  • Relevance of the proposed project to program priorities.

For information about NIH's commitment to supporting a safe and respectful work environment, who to contact with questions or concerns, and what NIH's expectations are for institutions and the individuals supported on NIH-funded awards, please see https://grants.nih.gov/grants/policy/harassment.htm .
  • Recipients of FFA must also administer their programs in compliance with applicable federal religious nondiscrimination laws and applicable federal conscience protection and associated anti-discrimination laws. Collectively, these laws prohibit exclusion, adverse treatment, coercion, or other discrimination against persons or entities on the basis of their consciences, religious beliefs, or moral convictions. Please see https://www.hhs.gov/conscience/conscience-protections/index.html and https://www.hhs.gov/conscience/religious-freedom/index.html .

Please contact the HHS Office for Civil Rights for more information about obligations and prohibitions under federal civil rights laws at https://www.hhs.gov/ocr/about-us/contact-us/index.html or call 1-800-368-1019 or TDD 1-800-537-7697.

In accordance with the statutory provisions contained in Section 872 of the Duncan Hunter National Defense Authorization Act of Fiscal Year 2009 (Public Law 110-417), NIH awards will be subject to the Federal Awardee Performance and Integrity Information System (FAPIIS) requirements. FAPIIS requires Federal award making officials to review and consider information about an applicant in the designated integrity and performance system (currently FAPIIS) prior to making an award. An applicant, at its option, may review information in the designated integrity and performance systems accessible through FAPIIS and comment on any information about itself that a Federal agency previously entered and is currently in FAPIIS. The Federal awarding agency will consider any comments by the applicant, in addition to other information in FAPIIS, in making a judgement about the applicant’s integrity, business ethics, and record of performance under Federal awards when completing the review of risk posed by applicants as described in 45 CFR Part 75.205 Federal awarding agency review of risk posed by applicants. This provision will apply to all NIH grants and cooperative agreements except fellowships.

Data Management and Sharing

Note: The NIH Policy for Data Management and Sharing is effective for due dates on or after January 25, 2023.

Consistent with the NIH Policy for Data Management and Sharing, when data management and sharing is applicable to the award, recipients will be required to adhere to the Data Management and Sharing requirements as outlined in the NIH Grants Policy Statement . Upon the approval of a Data Management and Sharing Plan, it is required for recipients to implement the plan as described.

3. Reporting

A final RPPR, invention statement, and the expenditure data portion of the Federal Financial Report are required for closeout of an award, as described in the NIH Grants Policy Statement .

The Federal Funding Accountability and Transparency Act of 2006 (Transparency Act), includes a requirement for awardees of Federal grants to report information about first-tier subawards and executive compensation under Federal assistance awards issued in FY2011 or later. All awardees of applicable NIH grants and cooperative agreements are required to report to the Federal Subaward Reporting System (FSRS) available at www.fsrs.gov on all subawards over $25,000.
